Several vaccines have been designed based on natural or designed protein scaffolds, which exhibited a potent adaptive immune response to antigens; however, antibodies are also generated against the scaffold, which may impair subsequent vaccination. In order to compare polypeptide scaffolds of different size and oligomerization state with respect to their efficiency, including anti-scaffold immunity, we compared several strategies of presentation of the RBD domain of the SARS-CoV-2 spike protein, an antigen aiming to generate neutralizing antibodies. A comparison of several genetic fusions of RBD to different nanoscaffolding domains (foldon, ferritin, lumazine synthase, and β-annulus peptide) delivered as DNA plasmids demonstrated a strongly augmented immune response, with high titers of neutralizing antibodies and a robust T-cell response in mice. Antibody titers and virus neutralization were most potently enhanced by fusion to the small β-annulus peptide scaffold, which itself triggered a minimal response in contrast to larger scaffolds. The β-annulus fused RBD protein increased residence in lymph nodes and triggered the most potent viral neutralization in immunization by a recombinant protein. Results of the study support the use of a nanoscaffolding platform using the β-annulus peptide for vaccine design. The results showed that the most significant impacts were the drop in the number of customers, which reduced the company’s income when compared to the values sold before the pandemic, and that the pandemic scenario generated a 45.3% reduction in financial transactions on average. In this research, we propose methods for technical condition assessment by applying the correlation of user-engagement score and Service Level Indicators (SLIs)/Service Level Objectives (SLOs)/Service Level Agreements (SLAs) measurements to identify user satisfaction types along with the infrastructure state. Our solution helps to improve content quality and, mainly, detect abnormal system behavior and poor infrastructure conditions. A straightforward interpretation of potential performance bottlenecks and vulnerabilities is achieved with the developed contingency table and correlation matrix for that purpose. We identify big data and system logs and metrics as the central sources that have performance issues during web-project usage. Throughout the analysis of an educational platform dataset, we found the main features of web-project content that have high user-engagement and provide value to services’ customers. According to our study, the usage and correlation of SLOs/SLAs with other critical metrics, such as user satisfaction or engagement improves early indication of potential system issues and avoids having users face them. These findings correspond to the concepts of SRE that focus on maintaining high service availability. In the German research project AVACON (AdVAnced Aircraft CONcepts), the boarding procedure is assessed using the PAXelerate boarding simulation. As the project demands an increased level of detail concerning the passenger movement model, this publication introduces an improved methodology. Additions to the model include the development of a method capable of describing the passenger walking speed in dependence of the surrounding objects, their proximity as well as the location of other passengers within the cabin. The validation of the model is performed using the AVACON research baseline and an Airbus A320. The model is then applied to an altered version of the Airbus A320 with an extended aisle and to a COVID-19 safe distance scenario. Regarding the results, an extended aisle width delivers boarding times reduced by up to 3%, whereas the COVID-19 assessment delivers a 67% increase in boarding times. Its course in many features resembles that of acute rheumatic fever (ARF). Therefore, it is interesting that the experiences with ARF can be used in the management of patients with PIMS-TS. The aim of the article is to analyse the current data on PIMS-TS in relation to ARF. PIMS-TS and ARF are associated with an abnormal immune response to specific pathogens (SARS-CoV-2 and group A streptococcus, respectively). The main symptoms of both diseases are fever and cardiac involvement. This study provides a framework for molecular characterization of novel VOC and investigates the effect of mutations on the binding affinity of the receptor-binding domain (RBD) to human angiotensin-converting enzyme 2 (hACE2) using in silico approach. Notable nonsynonymous mutations in RBD of VOC include the E484K and K417N/T that can be seen in South African and Brazilian variants, and N501Y and D614G that can be seen in all VOC. Phylogenetic analyses demonstrated that although the UK-VOC and the BR-VOC fell in the clade GR, they have different mutation signatures, implying an independent evolutionary pathway. The same is true about SA-VOC and COH-VOC felling in clade GH, but different mutation signatures. Combining molecular interaction modeling and the free energy of binding (FEB) calculations for VOC, it can be assumed that the mutation N501Y has the highest binding affinity in RBD for all VOC, followed by E484K (only for BR-VOC), which favors the formation of a stable complex. However, mutations at the residue K417N/T are shown to reduce the binding affinity. Once vaccination has started, there will be selective pressure that would be in favor of emergence of novel variants capable of escaping the immune system. Therefore, genomic surveillance should be enhanced to find and monitor new emerging SARS-CoV-2 variants before they become a public health concern.
