In this scenario, the drug repurposing approach is widely used for the rapid identification of potential drugs against SARS-CoV-2, considering viral and host factors. Methods We adopted a host transcriptome-based drug repurposing strategy utilizing the publicly available high throughput gene expression data on SARS-CoV-2 and other respiratory infection viruses. Based on the consistency in expression status of host factors in different cell types and previous evidence reported in the literature, pro-viral factors of SARS-CoV-2 identified and subject to drug repurposing analysis based on DrugBank and Connectivity Map (CMap) using the web tool, CLUE. Results The upregulated pro-viral factors such as TYMP, PTGS2, C1S, CFB, IFI44, XAF1, CXCL2, and CXCL3 were identified in early infection models of SARS-CoV-2. By further analysis of the drug-perturbed expression profiles in the connectivity map, 27 drugs that can reverse the expression of pro-viral factors were identified, and importantly, twelve of them reported to have anti-viral activity. The direct inhibition of the PTGS2 gene product can be considered as another therapeutic strategy for SARS-CoV-2 infection and could suggest six approved PTGS2 inhibitor drugs for the treatment of COVID-19. The computational study could propose candidate repurposable drugs against COVID-19, and further experimental studies are required for validation. India responded by drawing up National Containment Plan for COVID-19 that envisaged screening of people for COVID-19 for early detection of infection, providing community, primary health care-centered management for those with mild infection, tertiary care for those with severe infection, and health education to the population to reduce person-to-person transmission of infection. To minimize the spread of infection from other countries, all travelers were screened and quarantined; in the third week of March, India imposed ban on all international travel. To prevent person-to-person spread, congregation of people for commercial, educational, entertainment, sports, religious, and other social activities was banned. To minimize intracity, interstate, and urban rural spread, India imposed nationwide lockdown on 25th March 2020. The lockdown flattened the epidemic curve and provided needed time for the country to reorganize the health system, so that it can provide needed care for the increasing number of COVID-19 patients at all levels of health care. National guidelines for screening of people for severe acute respiratory syndrome-coronavirus-2 infection, quarantine, and home isolation and care of infected people were drawn up and implemented. Health education on methods by which person-to-person transmission of infection can be minimized has been provided using all media of communication. Global comparison shows that currently India has low infection rates, low fatality rates per 1,00,000, and high recovery rates. There are ongoing research studies to document the course and outcome of COVID-19 in India. Clinical trials of potential drugs for treatment of severe infections and vaccines for preventing infection in the high-risk groups are underway. A COVID-19 disease surveillance program is underway to track the infection rates and bring appropriate midcourse corrections in interventions. In the coming months, the people and health system will not only have to evolve a sustainable strategy to provide needed care for COVID-19 cases but also ensure that all other health care programs are implemented and patients with other illnesses get optimal care too. The restrictions can limit the movement of temporary labour that is already on the national territory and can prevent the arrival of other foreign workers to be engaged mainly in the harvesting of crops. In Italy, temporary workers support diverse farming activities and are mainly young immigrants engaged in working relationships characterized by extreme flexibility. They must be available for very different tasks, interacting with multiple farms and moving between different areas of the country, generally based on the succession of the harvesting calendars of the main crops. Reducing the mobility of this workforce on the national territory decreases its availability in local labour markets with worrying damage to agricultural production. This paper analyses the possible impact of this labour availability reduction in a farming area of Southern Italy (Centre-West Sardinia), where various farm types operate, with diverse productive orientations. An economic model estimates this impact on production and income based on the structural characteristics of these farm types, among others own farm labour endowment, as well as on the technical requirements of the productive processes, including labour needs. The model considers the expectations of farmers on the temporary labour availability and identifies the possible adaptations that can be undertaken to better contrast the reduction generated by the constraints to the mobility of individuals in the COVID-19 crisis. These adjustments result in changes in income levels, as well as employment in the single farm types and the area as a whole. There is a strong, albeit very diversified, impact on the types of farms. Significant reductions in income are found in the types that heavily depend on temporary labour for certain crop operations, especially harvesting. These effects are generated despite these farms change their activities, expanding some crops to make better use of their family labour and permanent employees. The less profitable types of farm, therefore even less able to attract external resources, considerably worsen their conditions; this would aggravate the social and environmental balance of the areas in which they operate, which are also the most marginal in the study area. We previously identified a novel accessory protein NS7a encoded by PDCoV; however, the function of NS7a was not resolved. In this study, we demonstrated that PDCoV NS7a is an interferon antagonist. Overexpression of NS7a notably inhibited Sendai virus (SeV)-induced interferon-β (IFN-β) production and the activation of IRF3 rather than NF-κB. NS7a also inhibited IFN-β promoter activity induced by RIG-I, MDA5, MAVS, TBK1, and IKKε, which are key components of the RIG-I-like receptor (RLR) signaling pathway but not IRF3, the transcription factor downstream of TBK1/IKKε. Surprisingly, NS7a specifically interacts with IKKε but not with the closely related TBK1. Furthermore, NS7a interacts simultaneously with the kinase domain (KD) and the scaffold dimerization domain (SDD) of IKKε, competing with TRAF3, and IRF3 for binding to IKKε, leading to the reduction of RLR-mediated IFN-β production. The interactions of TRAF3-IKKε and IKKε-IRF3 are also attenuated in PDCoV-infected cells. Taken together, our results demonstrate that PDCoV NS7a inhibits IFN-β production by disrupting the association of IKKε with both TRAF3 and IRF3, revealing a new mechanism utilized by a PDCoV accessory protein to evade the host antiviral innate immune response.
