                <text>Due to their ability to inhibit viral DNA or RNA replication, nucleoside analogues have been used for decades as potent antiviral therapeutics. However, one of the major limitations of nucleoside analogues is the development of antiviral resistance. In that regard, flexible nucleoside analogues known as &amp;#8220;fleximers&amp;#8221; have garnered attention over the years due to their ability to survey different amino acids in enzyme binding sites, thus overcoming the potential development of antiviral resistance. Acyclic fleximers have previously demonstrated antiviral activity against numerous viruses including Middle East Respiratory Syndrome coronavirus (MERS-CoV), Ebola virus (EBOV), and, most recently, flaviviruses such as Dengue (DENV) and Yellow Fever Virus (YFV). Due to these interesting results, a Structure Activity Relationship (SAR) study was pursued in order to analyze the effect of the pyrimidine functional group and acyl protecting group on antiviral activity, cytotoxicity, and conformation. The results of those studies are presented herein.</text>

                <text>Vitiation refers to the condition where the oxygen concentration in the air is reduced due to the mix of dilution gas. The vitiation effects on a premixed methane flame were investigated on a swirl-stabilized gas turbine model combustor under atmospheric pressure. The main purpose is to analyze the combustion stability and CO emission performance in vitiated air and compare the results with the flame without vitiation. The N2, CO2, and H2O (steam) were used as the dilution gas. Measurements were conducted in a combustor inlet temperature of 384 K and 484 K. The equivalence ratio was varied from stoichiometric conditions to the LBO (Lean Blowout) limits where the flame was physically blown out from the combustor. The chemical kinetics calculation was performed with Chemkin software to analyze the vitiation effects on the flame reaction zone. Based on the calculation results, the changes in the temperature gradient, CO concentration, and active radicals across the flame reaction zone were identified. The time-averaged CH chemiluminescence images were recorded and the results indicated the features of the flame shape and location. The CH signal intensity provided the information about the heat-release zone in the combustor. The combustion LBO limits were measured and the vitiation of CO2 and H2O were found to have a stronger impact to elevate the LBO limits than N2. Near the LBO limits, the instability of the flame reaction was revealed by the high-speed chemiluminescence imaging and the results were analyzed by FFT (Fast Fourier Transfer). CO emission was measured with a water-cooled probe which is located at the exit of the combustor. The combustion vitiation has been found to have the compression effect on the operation range for low CO emission. However, this compression effect could be compensated by improving the combustor inlet temperature.</text>

                <text>Background. Acute respiratory viral infection (ARVI) is accompanied by a general intoxication syndrome and respiratory mucosa predominant damage. A significant number of viral nature pathogens from various nosological groups (influenza viruses, parainfluenza, adenoviruses, respiratory syncytial viruses, coronaviruses, picornaviruses, rhinoviruses, enteroviruses, herpesviruses, etc.) could cause development of ARVI at human respiratory tract. Methods of ethnoscience and non-traditional medicine can be used as part of integrated treatment and prevention of influenza and ARVI. With the help of anti-cold homeopathic remedies, people can be treated from the very beginning of the disease when an elevated temperature is observed and there are all signs of intoxication and inflammation.  Objective. Scientific justification for the safety and efficacy of a complex homeopathic formulation in tablet form (Aconitum napellus D6, Ammonium bromatum D4, Atropa belladonna D6, Bryonia D6, Cinchona pubescens D6, Echinacea D3, Hydrargyrum bicyanatum D8, Rhustoxicodendron D6) intended to enhance the natural immunity in influenza and respiratory diseases, to normalize the functional state of the immune system and upper respiratory tract.  Results. Most patients have acute febrile respiratory syndrome, and some population groups have an increased risk of complications in the form of severe illness or death. These groups include elderly people, very young people, and people with concomitant illnesses. Annually, every third inhabitant of the planet suffers from acute respiratory infections. We have conducted a scientific substantiation of the safety and efficacy of a complex homeopathic tablet formulation (Aconitum napellus D6, Ammonium bromatum D4, Atropa belladonna D6, Bryonia D6, Cinchona pubescens D6, Echinacea D3, Hydrargyrum bicyanatum D8, Rhustoxico­dendron D6), which can be used to treat and prevention of these diseases. Conclusions. Components of developed homeopathic preparation are used in medical practice as anti-cold and immunomodulating mono- and complex homeopathic preparations for more than 100 years. There are no restrictions on the use of the drug in the context of its safety profile, since the active ingredient concentrations used are completely non-toxic. The preparation enhances the body’s protective responses and promotes the relief of symptoms of acute respiratory infections and flu (headache, sneezing, runny nose, sore throat, body aches, fever) and rapid recovery.</text>

                <text>First isolated in 1947 from a monkey in the Zika forest in Uganda, and from mosquitoes in the same forest the following year, Zika virus has gained international attention due to concerns for infection in pregnant women potentially causing fetal microcephaly. More than one million people have been infected since the appearance of the virus in Brazil in 2015. Approximately 80% of infected patients are asymptomatic. An association with microcephaly and other birth defects as well as Guillain-Barre Syndrome has led to a World Health Organization declaration of Zika virus as a Public Health Emergency of International Concern in February 2016. Zika virus is a vector-borne disease transmitted primarily by the Aedes aegypti mosquito. Male to female sexual transmission has been reported and there is potential for transmission via blood transfusions. After an incubation period of 2-7 days, symptomatic patients develop rapid onset fever, maculopapular rash, arthralgia, and conjunctivitis, often associated with headache and myalgias. Emergency department (ED) personnel must be prepared to address concerns from patients presenting with symptoms consistent with acute Zika virus infection, especially those who are pregnant or planning travel to Zika-endemic regions, as well as those women planning to become pregnant and their partners. The identify-isolate-inform (3I) tool, originally conceived for initial detection and management of Ebola virus disease patients in the ED, and later adjusted for measles and Middle East Respiratory Syndrome, can be adapted for real-time use for any emerging infectious disease. This paper reports a modification of the 3I tool for initial detection and management of patients under investigation for Zika virus. Following an assessment of epidemiologic risk, including travel to countries with mosquitoes that transmit Zika virus, patients are further investigated if clinically indicated. If after a rapid evaluation, Zika or other arthropod-borne diseases are the only concern, isolation (contact, droplet, airborne) is unnecessary. Zika is a reportable disease and thus appropriate health authorities must be notified. The modified 3I tool will facilitate rapid analysis and triggering of appropriate actions for patients presenting to the ED at risk for Zika.</text>

                <text>Toll-like Receptors (TLRs) sense viral infections and induce production of type I interferons (IFNs), other cytokines, and chemokines. Viral recognition by TLRs and other pattern recognition receptors (PRRs) has been proven to be cell-type specific. Triggering of TLRs with selected ligands can be beneficial against some viral infections. Macrophages are antigen-presenting cells that express TLRs and have a key role in the innate and adaptive immunity against viruses. Coronaviruses (CoVs) are single-stranded, positive-sense RNA viruses that cause acute and chronic infections and can productively infect macrophages. Investigation of the interplay between CoVs and PRRs is in its infancy. We assessed the effect of triggering TLR2, TLR3, TLR4, and TLR7 with selected ligands on the susceptibility of the J774A.1 macrophage cell line to infection with murine coronavirus (mouse hepatitis virus, [MHV]). Stimulation of TLR2, TLR4, or TLR7 did not affect MHV production. In contrast, pre-stimulation of TLR3 with polyinosinic-polycytidylic acid (poly I:C) hindered MHV infection through induction of IFN-β in macrophages. We demonstrate that activation of TLR3 with the synthetic ligand poly I:C mediates antiviral immunity that diminishes (MHV-A59) or suppresses (MHV-JHM, MHV-3) virus production in macrophages.</text>

                <text>Abstract Background Several lines of evidence indicate that memory loss represents a synaptic failure caused by soluble amyloid β (Aβ) oligomers. However, the pathological relevance of Aβ oligomers (AβOs) as the trigger of synaptic or neuronal degeneration, and the possible mechanism underlying the neurotoxic action of endogenous AβOs remain to be determined. Results To specifically target toxic AβOs in vivo, monoclonal antibodies (1A9 and 2C3) specific to them were generated using a novel design method. 1A9 and 2C3 specifically recognize soluble AβOs larger than 35-mers and pentamers on Blue native polyacrylamide gel electrophoresis, respectively. Biophysical and structural analysis by atomic force microscopy (AFM) revealed that neurotoxic 1A9 and 2C3 oligomeric conformers displayed non-fibrilar, relatively spherical structure. Of note, such AβOs were taken up by neuroblastoma (SH-SY5Y) cell, resulted in neuronal death. In humans, immunohistochemical analysis employing 1A9 or 2C3 revealed that 1A9 and 2C3 stain intraneuronal granules accumulated in the perikaryon of pyramidal neurons and some diffuse plaques. Fluoro Jade-B binding assay also revealed 1A9- or 2C3-stained neurons, indicating their impending degeneration. In a long-term low-dose prophylactic trial using active 1A9 or 2C3 antibody, we found that passive immunization protected a mouse model of Alzheimer's disease (AD) from memory deficits, synaptic degeneration, promotion of intraneuronal AβOs, and neuronal degeneration. Because the primary antitoxic action of 1A9 and 2C3 occurs outside neurons, our results suggest that extracellular AβOs initiate the AD toxic process and intraneuronal AβOs may worsen neuronal degeneration and memory loss. Conclusion Now, we have evidence that HMW-AβOs are among the earliest manifestation of the AD toxic process in mice and humans. We are certain that our studies move us closer to our goal of finding a therapeutic target and/or confirming the relevance of our therapeutic strategy.</text>

                <text>Species of Chydoridae provide the main diversity of the Cladocera. These organisms have been the subject of many studies; some dealing with their role in energy flow in aquatic ecosystems, since they inhabit the littoral region of water bodies which undergo the first impacts from anthropic activities. The aim of this study is to increase knowledge about the life cycle of Coronatella rectangula (Sars, 1861), a species found in several water bodies in the state of Minas Gerais, Brazil. The life cycle was determined by the culture of parthenogenetic females under controlled conditions in the laboratory. Experimental cultures were maintained in growth chambers at a constant temperature of 23.6(±0.5)ºC, through a 12 h light/12 h dark photoperiod. The organisms were fed on a suspension of Pseudokirchneriella subcapitata (Chlorophyceae) (10(5) cells.mL-1), and 0.02 mL of a mixed suspension of yeast and fish ration added per organism in equal proportions (1:1). Fifty parthenogenetic females with eggs were isolated and maintained until they produced neonates. Thirty of these neonates that had less than 24 hours were put in polypropylene bottles of 50 mL and kept in a germination chamber. These organisms were observed daily to obtain the parameters of the life cycle. Biomass and secondary production were also calculated. The embryonic development time of the specimens of C. rectangula was 1.68(±0.13) days and the time to reach primipara, was 2.48(±0.45) days. The mean fecundity of C. rectangula was two eggs/female/brood and the total number of eggs produced by the female during its life cycle was 27.8 eggs. During the whole life cycle, specimens of C. rectangula had a maximum of 14 seedlings, with two instars in the juvenile stage. Total biomass for C. rectangula was 36.66 µgDW.m-3(9.83 for the juvenile stage and 26.82 µgDW.m-3 for adults), and secondary production was 12.10 µgDW.m-3.day-1(8.34 µgDW.m-3.day-1 for egg production and 3.76 µgDW.m-3.day-1 for the juvenile stage).</text>

                <text>Abnormal iron metabolism is observed in many neurodegenerative diseases, however only two have shown dysregulation of brain iron homeostasis as the primary cause of neurodegeneration. Herein, we review one of these - hereditary ferritinopathy (HF) or neuroferritinopathy, which is an autosomal dominant, adult onset degenerative disease caused by mutations in the ferritin light chain (FTL) gene. HF has a clinical phenotype characterized by a progressive movement disorder, behavioral disturbances, and cognitive impairment. The main pathologic findings are cystic cavitation of the basal ganglia, the presence of ferritin inclusion bodies (IBs), and substantial iron deposition. Mutant FTL subunits have altered sequence and length but assemble into soluble 24-mers that are ultrastructurally indistinguishable from those of the wild type. Crystallography shows substantial localized disruption of the normally tiny 4-fold pores between the ferritin subunits because of unraveling of the C-termini into multiple polypeptide conformations. This structural alteration causes attenuated net iron incorporation leading to cellular iron mishandling, ferritin aggregation, and oxidative damage at physiological concentrations of iron and ascorbate. A transgenic murine model parallels several features of HF, including a progressive neurological phenotype, ferritin IB formation, and misregulation of iron metabolism. These studies provide a working hypothesis for the pathogenesis of HF by implicating (1) a loss of normal ferritin function that triggers iron accumulation and overproduction of ferritin polypeptides, and (2) a gain of a toxic function through radical production, ferritin aggregation, and oxidative stress. Importantly, the finding that ferritin aggregation can be reversed by iron chelators and oxidative damage can be inhibited by radical trapping may be used for clinical investigation. This work provides new insights into the role of abnormal iron metabolism in neurodegeneration.</text>

Related details of laboratory-confirmed MERS-CoV were extracted and analyzed by standard statistical methods.Results: A total of 107 cases of MERS-CoV, including 18 deaths (overall case fatality rate (CFR), 16.8%; male-specific CFR was 17.5% [14/80] and female-specific CFR was 14.8% [4/27]) were reported to WHO from the National International Health Regulation Focal Points of Saudi Arabia and Oman. The overall mean age was 50&amp;plusmn;17 years and 80 patients (74.8%) were male. The average time from the onset of the symptoms to the first hospitalization was 3&amp;plusmn;3.3 days; from the first hospitalization to laboratory confirmation was 3.6&amp;plusmn;6.5 days; from the onset of symptom to death was 17.5&amp;plusmn;11.7 days; and the mean length of hospitalization for&amp;nbsp;patients with MERS-CoV was 3.5&amp;plusmn;3.9 days. Males in comparison to females had a 1.5-fold increased chance (adjusted OR =1.5 [95% CI: 1.3&amp;ndash;1.8]) of death related to MERS-CoV infection; 1.05 [95% CI: 1.1&amp;ndash;3.3], 1.05 [95% CI: 1.2&amp;ndash;2.8] and 1.06 [95% CI: 1.2&amp;ndash;2.0] for those who had exposure to camels, camel milk consumption, and close contact with MERS-CoV cases, respectively.
