La zona de Cuba con mayor cantidad de ejemplares es la Norcentral (Archipiélago SabanaCamagüey) y la de menor fue la zona Suroccidental (Península de Guanahacabibes). Las zonas Suroriental, Noroccidental (Norte HabanaMatanzas) y Nororiental (Costa Norte de Oriente) presentaron una mayor proporción de algas pardas y el resto presentó una mayor proporción de algas verdes. ABSTRACT This study aimed to evaluate the presence of Cuban marine phycoflora in major herbaria in Cuba and determine what areas of the Cuban archipelago are better represented. To achieve the main Cuban herbal hosting macroalgae were visited, and all herbal materials were reviewed. According to these materials, in 12 collections of 11 national institutions, orders with more species of Rhodophyta were Compsopogonales, Nemaliales, Bonnemaisoniales, Rhodogorgonales and Gigartinales; for heterokonts, Fucales and Dictyotales; of Chlorophyta, Bryopsidales and Dasycladales. The access and use of green areas were analyzed using a survey conducted after the official COVID-19 emergency in Japan (16 April to 14 May, 2020). Visits to green areas during the pandemic have gained salience globally from multiple perspectives: health, planning, social justice, and equity. The results of this study demonstrated that socioeconomic factors influenced the frequency of visiting green areas. The factors further influenced the use of the three categories of green areas (parks, agricultural lands, and gardens). Environmental contexts, including the land use patterns in residential areas, also influenced the use of specific types of green areas. Thus, policies need to further facilitate visits to green areas by reflecting the socioeconomic attributes of residents and their households, including income, number of children, gender, and age, incorporating those who have less access and considering the spread of COVID-19 locally. Furthermore, policies for the use of specific green areas, including parks, agricultural lands, and gardens, need to take cognizance of the residents’ environmental contexts. The preparation and purification of native viral antigens in conventional cell-based protein expression systems are often accompanied by technical hardships. These challenges are attributable mainly to protein aggregation and insolubility during expression and purification, as well as to very low expression levels associated with the toxicity of some viral proteins. Here, we describe a novel approach for the production of monoclonal antibodies (mAbs) against nucleocapsid protein (NP) of the Middle East respiratory syndrome coronavirus (MERS-CoV). Using a wheat germ cell-free protein synthesis system, we successfully prepared large amounts of MERS-CoV NP antigen in a state that was highly soluble and intact for immunization. Following mouse immunization and hybridoma generation, we selected seven hybridoma clones that produced mAbs with exclusive reactivity against MERS-CoV NP. Epitope mapping and subsequent bioinformatic analysis revealed that these mAbs recognized epitopes located within relatively highly conserved regions of the MERS-CoV amino-acid sequence. Consistently, the mAbs exhibited no obvious cross-reactivity with NPs derived from other related viruses, including SARS coronavirus. After determining the optimal combinations of these mAbs, we developed an enzyme-linked immunosorbent assay and a rapid immunochromatographic antigen detection test that can be reliably used for laboratory diagnosis of MERS-CoV. Thus, this study provides strong evidence that the wheat germ cell-free system is useful for the production of diagnostic mAbs against emerging pathogens. Previously, we established a human DPP4 transgenic (hDPP4-Tg) mouse model in which we studied complement overactivation-induced immunopathogenesis. Here, to better understand the pathogenesis of MERS-CoV, we studied the role of pyroptosis in THP-1 cells and hDPP4 Tg mice with MERS-CoV infection. We found that MERS-CoV infection induced pyroptosis and over-activation of complement in human macrophages. The hDPP4-Tg mice infected with MERS-CoV overexpressed caspase-1 in the spleen and showed high IL-1&beta; levels in serum, suggesting that pyroptosis occurred after infection. However, when the C5a-C5aR1 axis was blocked by an anti-C5aR1 antibody (Ab), expression of caspase-1 and IL-1&beta; fell. These data indicate that MERS-CoV infection induces overactivation of complement, which may contribute to pyroptosis and inflammation. Pyroptosis and inflammation were suppressed by inhibiting C5aR1. These results will further our understanding of the pathogenesis of MERS-CoV infection. To improve the detection rate of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), we analyzed the results of viral nucleic acid and serum-specific antibody tests on clinical samples from 20 patients with SARS-CoV-2 infection diagnosed at the First Affiliated Hospital of Guangzhou Medical University in China. By comparing various sample types collected from COVID-19 patients, we revealed multiple pathways for SARS-CoV-2 shedding, and a prolonged detectable period for viral nucleic acid test in sputum specimens, demonstrating that the timeline of the viral shedding is of great value in determining the time of release from quarantine or discharge from hospital. We also recommend for the application of serological test to assist in confirming SARS-CoV-2 infection judged by viral nucleic acid test, especially when COVID-19-related symptoms have appeared and the viral nucleic acid test was negative. Our findings are critical for the diagnosis of SARS-CoV-2 infection and for determining deadline of restriction measures to prevent transmission caused by convalescent patients with COVID-19.
