Most previous structural studies on S protein have been conducted at pH 8.0, but knowledge of the conformational propensities under both physiological and endosomal pH conditions is important to inform vaccine development. Our current study employed single-particle cryoelectron microscopy to visualize multiple states of open and closed conformations of S protein at physiological pH 7.4 and near-physiological pH 6.5 and pH 8.0. Propensities of open and closed conformations were found to differ with pH changes, whereby around 68% of S protein exists in open conformation at pH 7.4. Furthermore, we noticed a continuous movement in the N-terminal domain, receptor-binding domain (RBD), S2 domain, and stalk domain of S protein conformations at various pH values. Several key residues involving RBD-neutralizing epitopes are differentially exposed in each conformation. This study will assist in developing novel therapeutic measures against SARS-CoV2. Previous structural studies of membrane fusion proteins have largely used crystallography of static molecular fragments, in isolation of their transmembrane domains. In this study we have produced purified, irradiated SARS-CoV virions that retain their morphology, and are fusogenic in cell culture. We used cryo-electron microscopy and image processing to investigate conformational changes that occur in the entire spike of intact virions when they bind to the viral receptor, angiotensin-converting enzyme 2 (ACE2). We have shown that ACE2 binding results in structural changes that appear to be the initial step in viral membrane fusion, and precisely localized the receptor-binding and fusion core domains within the entire spike. Furthermore, our results show that receptor binding and subsequent membrane fusion are distinct steps, and that each spike can bind up to three ACE2 molecules. The SARS-CoV spike provides an ideal model system to study receptor binding and membrane fusion in the native state, employing cryo-electron microscopy and single-particle image analysis. Objetivou-se avaliar a qualidade do leite produzido em unidades de produção familiares, no sul do Espírito Santo, para verificar o atendimento aos padrões legais. Foram realizadas três coletas em 29 tanques comunitários totalizando 87 amostras. Realizaram-se análises de contagem padrão em placa (CPP), contagem de bactérias psicrotróficas (CBP), contagem de células somáticas (CCS), acidez titulável, densidade, gordura, extrato seco total (EST), extrato seco desengordurado (ESD), pesquisa de resíduos de antibióticos (β-lactâmicos e tetraciclina) e presença de cádmio e chumbo. Das 87 amostras, 66% apresentaram valores fora do padrão para CPP e 38% para CCS. Quanto às análises físico-químicas, 85% das amostras apresentaram resultados fora do padrão para ESD, 10,3% para densidade e 2,3% para acidez titulável. Todas as amostras atenderam a legislação quanto a presença de resíduos de antibióticos, teores de cádmio e chumbo. Agricultores familiares, ao terem consciência do potencial dessas plantas alimentícias, podem diversificar sua alimentação habitual e cultivá-las aproveitando áreas improdutivas ou de baixa fertilidade. Com isso, objetivou-se avaliar o nível de conhecimento de agricultores moradores em assentamentos rurais sobre as PANC, identificando as espécies presentes nos locais estudados. A pesquisa foi realizada entre março e junho de 2019 em três assentamentos da reforma agrária de Santana do Livramento, Rio Grade do Sul, com 15 informantes-chave, utilizando como metodologia a amostragem tipo bola de neve. Perguntas pré-elaboradas foram respondidas pelos agricultores assentados durante o método da turnê guiada. Como resultados, foram identificadas 20 espécies de PANC pertencendo a 12 famílias botânicas, sendo a maioria da família Astereaceae (38,8%), localizadas principalmente em pastagens naturais (50%). As espécies de maior ocorrência nos assentamentos foram a carqueja (Baccharis trimera), a buva (Conyza bonariensis), o picão preto (Bidens pilosa), a beldroega (Portulaca oleracea), as folhas da abóbora moranga (Curcubita maxima) e o caruru (Amaranthus deflexus). Observou-se que o conhecimento dos agricultores acerca das PANC é reduzido e baseado em crenças individuais e valores ancestrais, o que refletiu na ausência do consumo das espécies vegetais. In these urban homegardens, there are plants and also people who have familiarity with these plants, which allow us to consider these spaces as holders of botanical knowledge on food plants. Cultural and socioeconomic changes have warned that homegardens and botanical knowledge held by this population have become vulnerable. The present paper aims to discuss the knowledge about plants and the relevance of the maintenance of cultivated homegardens for the permanence and transmission of this botanical knowledge, here considered as heritage. In order to understand the richness and vulnerability of this group, residents who had urban homegardens with food cultivation plants were interviewed in order to characterize the homegardens and understand their motivations for food cultivation. From the recommended procedures for the understanding of urban ethnobotany, the interviews allowed us to understand the actions required for the maintenance of this heritage’s existence, which results in measures to contain its vulnerabilities. O objetivo do trabalho foi o de avaliar o conhecimento dos consumidores a respeito dos conceitos relacionados a selos de qualidade em Dracena/SP. Para isso, foram feitas 257 entrevistas pessoais com o auxílio de questionário estruturado, entre abril e junho de 2018. Os resultados indicam que 1/3 dos consumidores desconhecem a agricultura orgânica e pouco mais da metade desconhecem os conceitos da agricultura familiar. Os produtos de quilombos e de comércio justo são praticamente desconhecidos em Dracena. Verificou-se uma relação positiva entre os níveis de estudos e o conhecimento da agricultura familiar e produtos orgânicos. Como conclusão, o trabalho mostrou que é preciso realizar um amplo e contínuo trabalho de informação dos consumidores sobre os selos de qualidade. Trata-se de um estudo transversal misto, realizado em 2014 em São José de Ubá, RJ. A pesquisa é composta por uma etapa qualitativa, baseada em observações em campo e entrevistas com 25 participantes sobre CAP, no que se refere ao uso de agrotóxicos. A etapa quantitativa abrange avaliação sociodemográfica e de exposição aos agrotóxicos de 78 agricultores. Apesar de reconhecerem parcialmente os perigos dos agrotóxicos, os agricultores enfrentam os riscos da exposição e comumente adotam práticas laborais inadequadas. Fatores como a carência de apoio técnico e treinamento laboral, baixa escolaridade, dificuldade de compreensão das orientações de bulas e rótulos dos agrotóxicos, valor elevado e desconforto causado pelo equipamento de proteção individual (EPI), além da crença da dependência do uso de agrotóxicos, explicam parcialmente as atitudes não seguras. É preciso promover práticas agrícolas mais sustentáveis, fortalecer o apoio técnico e treinamento laboral, melhorar a comunicação e o gerenciamento dos riscos e reduzir as desigualdades de gênero entre os agricultores brasileiros, como a forma mais eficaz e imediata de prevenir os agravos relacionados ao trabalho com agrotóxicos.
