The pathogenesis and host immune responses of PEAV infection are not fully characterized. The reason lies in the stomach environment, which would degrade cell-cultured live viruses via oral infection, making it difficult to establish an effective infection model to study the pathogenesis and host immune responses in pigs with a mature immune system. To solve this problem, in this study, coated PEAV-loaded microspheres were developed by centrifugal granulation-fluidized bed coating and demonstrated as an effective oral delivery system/animal infection model to protect PEAV virion against the complex gastrointestinal environment in vitro and to cause infection in weaned piglets in vivo. Weaned piglets orally inoculated with coated PEAV-loaded microspheres developed diarrhea and virus RNA was detected in rectal swabs from one to seven days post inoculation. In addition, microscopic lesions in the small intestine were observed, and viral antigens were also detected in the small intestines with PEAV immunohistochemical staining. Importantly, PEAV significantly inhibited mRNA expression of IFN-α, IFN-β, OAS, Mx1, and PKR, the genes involved in modulation of the host immune responses, in infected Peyer's patches, indicating that PEAV can overcome the antiviral response to cause damage when infection occurs. Collectively, our research successfully established a PEAV animal infection model in weaned piglets and suggested that the observed gene expression profile might help explain immunological changes associated with PEAV infection. Like many other human and animal viruses, coronavirus infection of mammalian cells induces apoptosis. In this study, the global gene expression profiles are first determined in IBV-infected Vero cells at 24 hours post-infection by Affymetrix array, using avian coronavirus infectious bronchitis virus (IBV) as a model system. It reveals an up-regulation at the transcriptional level of both pro-apoptotic Bak and pro-survival myeloid cell leukemia-1 (Mcl-1). These results were further confirmed both in vivo and in vitro, in IBV-infected embryonated chicken eggs, chicken fibroblast cells and mammalian cells at transcriptional and translational levels, respectively. Interestingly, the onset of apoptosis occurred earlier in IBV-infected mammalian cells silenced with short interfering RNA targeting Mcl-1 (siMcl-1), and was delayed in cells silenced with siBak. IBV progeny production and release were increased in infected Mcl-1 knockdown cells compared to similarly infected control cells, while the contrary was observed in infected Bak knockdown cells. Furthermore, IBV infection-induced up-regulation of GADD153 regulated the expression of Mcl-1. Inhibition of the mitogen-activated protein/extracellular signal-regulated kinase (MEK/ERK) and phosphoinositide 3-kinase (PI3K/Akt) signaling pathways by chemical inhibitors and knockdown of GADD153 by siRNA demonstrated the involvement of ER-stress response in regulation of IBV-induced Mcl-1 expression. A detailed scientific analysis of this pheno-menon is still to come, but now it is urgently needed to evaluate and compare the disease dynamics in order to im-prove the quarantine activities and the level of individual protection, to speed the rate of isolation of infected per-sons, etc. In mid-March 2020, the WHO reported a sharp exponential increase in the number of COVID-19 cases in the world. Therefore, the study of the dynamics of the pandemic continues to attract the interest of researchers. However, the receipt of new data requires constant analysis of the pandemic dynamics. Objective. In this paper we compare the pandemic dynamics in March and April, 2020 for different countries in Eu-rope, USA, regions and worldwide and try to estimate some global trends. Methods. We use official data about the accumulated numbers of confirmed COVID-19 cases in different countries and worldwide from WHO daily situation reports. In order to compare the epidemic dynamics in different countries, different time synchronization procedures and comparing with the exponential growth are used. Results. For Italy, Spain, Germany, Switzerland, France and USA, the epidemic dynamic in March, 2020 was com-pared with the situation in the Republic of Korea. The epidemic developments in Ukraine, Austria, their neighboring countries and global trends were analyzed. Conclusions. The situation with the COVID-19 pandemic is still threatening. But may be the dynamics in April, 2020 gives some hope for its stabilization at least in the countries with proper quarantine measures, fast detection and isolation of sick persons. The final sizes and durations of epidemic can be very different for different countries. Repeated outbreaks are possible and already occurring, but they will no longer be so severe, primarily because we have learned to better identify and isolate infected persons. The research set out to evaluate how the disaster emergency response was carried out amid the COVID-19 pandemic in the Special Region of Yogyakarta (SRY). The research employs qualitative observation of adaptive governance variables, i.e., infrastructure availability, information, conflict mechanism, regulation, and adaptation. The research analyzed primary data collected from focus group discussions with key persons at the Local Disaster Management Agency, Local Development Planning Agency, and Disaster Risk Reduction Platform responsible for the crisis and included an online survey to validate data. The research revealed that the SRY had exhibited adaptive governance to the COVID-19 pandemic, as apparent by, among others, open-access spatial and non-spatial data, extensive combined uses of both types of data, and prompt active engagement of communities in the enforcement of new rules and regulations mandated by national and provincial governments. Furthermore, during emergency responses to COVID-19, the stakeholders provided infrastructure and information, dealt with conflicts in multiple spatial units, encouraged adaptations, and formulated emergent rules and regulations. Materials and Methods: Time required for replenishing consumables and clinic preparedness to function under “new norms” of pandemic was documented. Infection mitigation measures and triaging strategy were evaluated. KPIs following resumption were analyzed. The Student's t-test was performed for comparing parameters. Results: Thirty percent of the patients consulted through telemedicine accepted or were eligible to initiate treatment on clinic resumption. Lack of safe transport and financial constraints prevented majority from undergoing IVF, and 9% delayed treatment due to fear of pandemic. With adequate training, staff compliance to meet new demands was achieved within a week, but procuring consumables and injections was time-consuming. Fifty-two cycles of IVF were performed including fresh and frozen embryo transfers with satisfactory KPIs even during pandemic. Conscious sedation and analgesia during oocyte retrieval were associated with reduced procedure time and no intervention for airway maintenance compared to general anesthesia. Self-reported pain scores by patients ranged from nil to mild on a graphic rating scale. Conclusions: This study provides practical insight for the resumption of IVF services during the COVID-19 pandemic. This research aimed to investigate the effectiveness of mE-Book as instructional content for Arabic language learning in Canvas LMS and the extent of mE-Book acceptance by students in the framework of the Technology Acceptance Model (TAM) 3. This research used a nonrandomized control group pretest-posttest design by comparing two groups with and without mE-Book and the interview to know the students’ point of view about mE-Book. The results showed that mE-Book was effective as instructional content for Arabic language learning in Canvas LMS with large effect based on effect size criteria. Students’ perspectives showed that the use of mE-Book as instructional content for Arabic language learning in Canvas LMS is interesting for online and offline learning. In conclusion, students could accept mE-Book in learning activity in Canvas LMS because it eased students in Arabic language learning.
