Además, se determinó el índice de fluidez para estimar su capacidad de procesamiento. De los ensayos realizados se obtuvieron polímeros estructuralmente semejantes al poliestireno, con un grado de contaminación menor al 1.6 % de agente precipitante, presentan un índice de fluidez entre 15.56 y 23.60 g/10 min por lo que pueden ser reprocesados por inyección y extrusión, la temperatura de transición vítrea y el peso molecular son similares a los residuos de EPS. El ensayo con mejores resultados fue la disolución 30 % EPS y precipitación con una relación volumétrica 1/3 THF/EG, ofreciendo un polímero sin contaminación por el etilenglicol remanente. Abstract: In this project the recycling process of expanded polystyrene (EPS) was studied by the method of dissolution - precipitation, as an alternative to minimize the environmental impact post-consumer. Thus, recycling of EPS packaging was developed varying the conditions of the method in order to recover polystyrene reprocessing. The EPS waste collected and fragmented were dissolved in tetrahydrofuran (THF) and precipitated with ethylene glycol (EG) under continuous mechanical stirring. The precipitate polymer was recovered with filtration, through drying, crushing and washing. The recovered polymers are structural characterized by the infrared spectroscopy, thermal by differential scanning calorimetry and thermogravimetry. Melt flow index was measured in order to estimate its processability. The tests performed indicate that the polymers obtained are structural identical to polystyrene with a lower degree of contamination to 1.6 % of precipitant, they have a melt index between 15.56 and 23.60 g/10 min so can be reprocessed by injection and extrusion, the glass transition temperature and molecular weight are similar to the waste EPS. The best performing test was 30 % EPS solution and precipitation with a volume ratio 1/3 THF/EG, offer a polymer free ethylene contamination. Mullins Description An account of the resource While human immunodeficiency virus type 1 (HIV-1)-specific cytotoxic T lymphocytes preferentially target specific regions of the viral proteome, HIV-1 features that contribute to immune recognition are not well understood. One hypothesis is that similarities between HIV and human proteins influence the host immune response, i.e., resemblance between viral and host peptides could preclude reactivity against certain HIV epitopes.We analyzed the extent of similarity between HIV-1 and the human proteome. Proteins from the HIV-1 B consensus sequence from 2001 were dissected into overlapping k-mers, which were then probed against a non-redundant database of the human proteome in order to identify segments of high similarity. We tested the relationship between HIV-1 similarity to host encoded peptides and immune recognition in HIV-infected individuals, and found that HIV immunogenicity could be partially modulated by the sequence similarity to the host proteome. ELISpot responses to peptides spanning the entire viral proteome evaluated in 314 individuals showed a trend indicating an inverse relationship between the similarity to the host proteome and the frequency of recognition. In addition, analysis of responses by a group of 30 HIV-infected individuals against 944 overlapping peptides representing a broad range of individual HIV-1B Nef variants, affirmed that the degree of similarity to the host was significantly lower for peptides with reactive epitopes than for those that were not recognized.Our results suggest that antigenic motifs that are scarcely represented in human proteins might represent more immunogenic CTL targets not selected against in the host. This observation could provide guidance in the design of more effective HIV immunogens, as sequences devoid of host-like features might afford superior immune reactivity. Expression of the S1 protein in the lungs and kidneys was detected using conventional polymerase chain reaction (PCR) and immunohistochemistry (IHC) targeting specific regions within the S1 subunit at weeks 3, 4, 5, and 6 after the first vaccination. Antigen-specific humoral and cellular immune responses were evaluated in serum and in cell culture following in vitro stimulation with a specific 9-mer epitope within the S1 protein (CYSSLILDY).Results: S1 protein expression was only detected by IHC in the kidneys of the Ad-MERS-S1 group at week 6 from first immunization, and in both lungs and kidneys of Ad-MERS-S1 group by conventional PCR at weeks 3 and 5 post-prime. The vaccine elicited a specific S1-immunoglobulin G antibody response, which was detected in the sera of the vaccinated mice at weeks 4 and 6 from the onset of the first immunization. There was a significant increase in the amount of Th1-related cytokines (interferon-γ and interleukin [IL] 12), and a significant decrease in the Th2-related cytokine IL-4 in splenocyte cell culture of the vaccinated group compared with the control groups.Conclusion: The results of this study suggest that this recombinant adenovirus vaccine encoding the S1 subunit of MERS-CoV elicits potentially protective antigen-specific humoral and cellular immune responses in mice. This virus (rTGEV-RS-SPEDV) was attenuated in highly-sensitive five-day-old piglets, as infected animals did not lose weight and none of them died. In addition, the virus caused very minor tissue damage compared with a virulent virus. The rTGEV-RS-SPEDV vaccine candidate was also attenuated in three-week-old animals that were used to evaluate the protection conferred by this virus, compared with the protection induced by infection with a virulent PEDV US strain (PEDV-NVSL). The rTGEV-RS-SPEDV virus protected against challenge with a virulent PEDV strain, reducing challenge virus titers in jejunum and leading to undetectable challenge virus RNA levels in feces. The rTGEV-RS-SPEDV virus induced a humoral immune response specific for PEDV, including neutralizing antibodies. Altogether, the data indicated that rTGEV-RS-SPEDV is a promising vaccine candidate against virulent PEDV infection.
