                <text>The interest in and use of 3D models in built environments is rapidly increasing, and they are now a key component of decision-making in areas including climate change mitigation (e.g., calculating solar panel potential, flood modelling, modelling housing age for retrofitting of thermal insulation), urban planning and cadastral systems (modelling rights, restrictions and responsibilities in complex buildings, streamlining the process to issue planning permits, design of existing or new developments) and infrastructure (construction, transport, utility management and modelling, asset management). 3D models are also an integrator for the data underpinning smart cities – knowing where a sensor is in 3D space allows the data to be integrated with the surrounding context – for example, noise data could be integrated with traffic information. Reflecting this interest, national mapping and cadastral agencies (NMCA) including Ordnance Survey (GB) are now increasingly generating 3D mapping at national scale, and there is extensive research as to how this data can be integrated with another emerging source of 3D models such as building information modelling (BIM).These trends were evident during the 3rd BIM/GIS Integration Workshop and 15th 3DGeoInfo 2020 events, which were co-hosted by University College London and Ordnance Survey (GB) in September 2020. The workshop and conference brought together international researchers from academia, industry, government and national mapping and cadastral agencies in the field of 3D geoinformation, in an interdisciplinary gathering of researchers in the fields of data collection, data management, data quality, data analysis, advanced modelling approaches, applications, users, visualisation, augmented reality, artificial intelligence and many more.This year’s theme was Users and Use Cases. The workshop and conference covered a wide range of topics including 3D data acquisition and processing, 3D city modelling and related standards, visualisation and dissemination of 3D data, augmented and virtual reality, 3D and Artificial Intelligence/Machine Learning. Three sessions of the BIM/GIS Integration Workshop were dedicated to Applications of BIM/GIS Integration, and an entire day of 3DGeoInfo 2020 to Users and Use Cases within 3DGeoInfo. Additionally, two sessions were specifically aimed at NMCA participants.Although initially intended to be a face-to-face event in London, the team rapidly adjusted to the emerging COVID-19 situation, identifying an online solution that facilitated and encouraged participant interaction. This meant that the events could still provide a platform for learning, discussion, and exchange of ideas that they have been able to in previous years, as well as providing opportunities to promote international collaboration in these topics. This special issue of the ISPRS International Annals of Photogrammetry, Remote Sensing and Spatial Information Sciences contains 22 papers selected by double-blind peer review carried out by a minimum of three reviewers. It includes research on topics including data modelling, indoor 3D, standards (IndoorML, CityGML, CityJSON), cadastral systems, pedestrian navigation, infrastructure, visualisation, point cloud management, data interoperability, data quality, generating national 3D datasets and planning/permit processes amongst others.</text>

                <text>The aim of this study is to examine the use of an inflammasome competitor as a preventative agent. Coronaviruses have zoonotic potential due to the adaptability of their S protein to bind receptors of other species, most notably demonstrated by SARS-CoV. The binding of SARS-CoV-2 to TLR (Toll-like receptor) causes the release of pro-IL-1β, which is cleaved by caspase-1, followed by the formation and activation of the inflammasome, which is a mediator of lung inflammation, fever, and fibrosis. The NLRP3 (NACHT, LRR and PYD domains-containing protein 3) inflammasome is implicated in a variety of human diseases including Alzheimer’s disease (AD), prion diseases, type 2 diabetes, and numerous infectious diseases. By examining the use of 4,4′-diaminodiphenyl sulfone (DDS) in the treatment of patients with Hansen’s disease, also diagnosed as Alzheimer’s disease, this study demonstrates the diverse mechanisms involved in the activation of inflammasomes. TLRs, due to genetic polymorphisms, can alter the immune response to a wide variety of microbial ligands, including viruses. In particular, TLR2Arg677Trp was reported to be exclusively present in Korean patients with lepromatous leprosy (LL). Previously, mutation of the intracellular domain of TLR2 has demonstrated its role in determining the susceptibility to LL, though LL was successfully treated using a combination of DDS with rifampicin and clofazimine. Of the three tested antibiotics, DDS was effective in the molecular regulation of NLRP3 inflammasome activators that are important in mild cognitive impairment (MCI), Parkinson’s disease (PD), and AD. The specific targeting of NLRP3 itself or up-/downstream factors of the NLRP3 inflammasome by DDS may be responsible for its observed preventive effects, functioning as a competitor.</text>

Although the compound has been studied extensively for its neuronal handling and synaptic actions, serotonin 5-HT receptors can be found extra-synaptically and not only in neurons but in many types of mammalian cells, inside and outside the central nervous system (CNS). Iniciamos con una discusión acerca de la influencia de las tecnologías y el aprendizaje informal en la educación formal. A partir de allí, exploramos las limitaciones de las teorías clásicas de aprendizaje para explicar el conocimiento generado en contextos de aprendizaje no estructurados. Abordamos el paradigma conectivista como estrategia para manejar estas limitaciones teóricas, y establecemos las bases de un modelo ontológico que permita su reconocimiento. Asimismo, usamos el paradigma de ecologías de conocimiento, para caracterizar un modelo de analítica de datos que permite desplegar el paradigma conectivista a partir de las interacciones que emergen en un proceso de aprendizaje. Finalmente, explicamos los detalles de su usopara un caso de estudio llamado SaCI, que es un ambiente inteligente de aprendizaje.</text>

Los sujetos de investigación, lo conformaron dieciséis (16) docentes, a los que se les suministró un cuestionario para la recolección de los datos con la técnica de la encuesta y analizándose por medio de la estadística descriptiva, generando así las conclusiones sobre la existencia de condiciones favorables a nivel conceptual y programático para el diseño del programa referido; generando una herramienta didáctica para a una futura propuesta curricular sobre Agroecología a ser impartida en la Carrera de Educación Agropecuaria.</text>

                <text>Abstract Background The RNA interference (RNAi) pathway is a mechanism of gene-suppression with potential gene therapy applications for treating viral disease such as HIV-1. The most suitable inducer of RNAi for this application is short hairpin RNA (shRNA) although it is limited to suppressing a single target. A successful anti-HIV-1 therapy will require combinations of multiple highly active, highly conserved shRNAs to adequately counter the emergence of resistant strains. Results We calculated the percentage conservations of 8, 846 unique 19 nucleotide HIV-1 targets amongst 37, 949 HIV-1 gene sequence fragments containing 24.8 million 19 mers. We developed a novel method of determining conservation in 'profile' sets of 5 overlapping 19 mer sequences (covering 23 nucleotides in total) to ensure that the intended conservation of each shRNA would be unaffected by possible variations in shRNA processing. Ninety six of the top ranking targets from 22 regions were selected based on conservation profiles, predicted activities, targets and specific nucleotide inclusion/exclusion criteria. We constructed 53 shRNAs with 20 bp stems and 43 shRNAs with 21 bp stems which we tested and ranked using fluorescent reporter and HIV-1 expression assays. Average suppressive activities ranged from 71 – 75%, with 65 hairpins classed as highly active (&gt; 75% activity). Overall we found little difference in activities from minor changes in stem length (20 cf. 21), or between neighboring targets differing by a single nucleotide in start position. However, there were several exceptions which suggest that all sequences, irrespective of similarities in target site or design, may be useful candidates. We encountered technical limitations with GFP reporter assays when the target domain was long and or when the distance between the target site and fusion junction was large. Assay performance was improved by dividing large targets into several shorter domains. Conclusion In summary, our novel selection process resulted in a large panel of highly active shRNAs spanning the HIV-1 genome, representing excellent candidates for use in multiple shRNA gene therapies. Our core selection method ensuring maximal conservation in the processed product(s) is also widely applicable to other shRNA applications.</text>

The nematode Caenorhabditis elegans is an important model for studying principles of gene regulation in multi-cellular organisms. A comprehensive parts list of putative regulatory motifs was yet missing for this model system. In this study, we compile a set of putative regulatory motifs by combining evidence from conservation and expression data. Description We present an unbiased comparative approach to a regulatory motif compendium for Caenorhabditis species. This involves the assembly of a new nematode genome, whole genome alignments and assessment of conserved k-mers counts. Candidate motifs are selected from a set of 9,500 randomly picked genes by three different motif discovery strategies. Motif candidates have to pass a conservation enrichment filter. Motif degeneracy and length are optimized. Retained motif descriptions are evaluated by expression data using a non-parametric test, which assesses expression changes due to the presence/absence of individual motifs. Finally, we also provide condition-specific motif ensembles by conditional tree analysis. Conclusion The nematode genomes align surprisingly well despite high neutral substitution rates. Our pipeline delivers motif sets by three alternative strategies. Each set contains less than 400 motifs, which are significantly conserved and correlated with 214 out of 270 tested gene expression conditions. This motif compendium is an entry point to comprehensive studies on nematode gene regulation. Visual analog scale (VAS) score, Harris score and Association Research Circulation Osseous (ARCO) stage of imaging examination were compared among those before treatment, and at 1, 3, 6, 9 and 12&amp;nbsp;years after treatment. Paired t-test was used for statistical analysis of VAS and Harris score before and after treatment. Difference of effective rate on all stages was analyzed with Chi-square test.Results: With 12-year follow-up, significant improvements on VAS (6.81 of pre-treatment vs 3.94 of 12-year post-treatment) and Harris score (74.54 of pre-treatment vs 80.14 of 12-year post-treatment) were observed (all p&amp;lt;0.05). Effective rate showed statistical significance among three stages of ARCO (p&amp;lt;0.05). The combined treatment showed different efficacies on different ARCO stages; the best was on ARCO Phase I.Conclusion: The combined treatment may delay or discontinue the development of ONFH in post-SARS patients.
