The choanoflagellate Salpingoeca rosetta expresses genes necessary for animal development and can alternate between unicellular and multicellular states, making it a powerful model for investigating the origin of animal multicellularity and mechanisms underlying cell differentiation. To compare the subcellular architecture of solitary collar cells in S. rosetta with that of multicellular 'rosette' colonies and collar cells in sponges, we reconstructed entire cells in 3D through transmission electron microscopy on serial ultrathin sections. Structural analysis of our 3D reconstructions revealed important differences between single and colonial choanoflagellate cells, with colonial cells exhibiting a more amoeboid morphology consistent with higher levels of macropinocytotic activity. Comparison of multiple reconstructed rosette colonies highlighted the variable nature of cell sizes, cell-cell contact networks, and colony arrangement. Importantly, we uncovered the presence of elongated cells in some rosette colonies that likely represent a distinct and differentiated cell type, pointing toward spatial cell differentiation. Intercellular bridges within choanoflagellate colonies displayed a variety of morphologies and connected some but not all neighbouring cells. Reconstruction of sponge choanocytes revealed ultrastructural commonalities but also differences in major organelle composition in comparison to choanoflagellates. Together, our comparative reconstructions uncover the architecture of cell differentiation in choanoflagellates and sponge choanocytes and constitute an important step in reconstructing the cell biology of the last common ancestor of animals. Coronavirus disease, caused by the severe acute respiratory syndrome coronavirus 2, is causing serious problems related to public health and socioeconomic status across the globe. Having started in Asia, the disease has already reached Europe, Africa, and more recently the Americas; hence, it is necessary to understand the context in which it spreads in regions that carry high risks associated with social vulnerability, as in the case of Latin America. Brazil is the South American country most affected by the pandemic—its first case was confirmed at the end of February 2020, and in less than two months, its total number of cases has already crossed 25,000. Among its worst-hit states, Ceará in Northeast Brazil worries health agencies as there have already been more than 2,000 cases in the 30 days of the outbreak in the state. Even more worrying is its capital Fortaleza, which has become the epicenter of the spread of the disease in Ceará, accounting for 86.5% cases and 67% deaths in the state—95% of the neighborhoods in Fortaleza have confirmed cases of the disease. An aspect that particularly causes concern is the disease’s diffusion, from neighborhoods with the highest family income to those with the lowest, directly affecting the socially vulnerable populations in territories with a majority of precarious settlements; in Fortaleza, favelas are the main type of precarious settlements (73.6%). Here we reported that SUPT16H, a subunit of FACT, is acetylated at lysine 674 (K674) of middle domain (MD), which involves TIP60 histone acetyltransferase. Such acetylation of SUPT16H is recognized by bromodomain protein BRD4, which promotes protein stability of SUPT16H. We further demonstrated that SUPT16H-BRD4 associates with histone modification enzymes (EZH2, HDAC1) and affects histone marks (H3K9me3, H3K27me3 and H3ac). BRD4 is known to profoundly regulate interferon (IFN) signaling, while such function of SUPT16H has never been explored. Surprisingly, our results revealed that SUPT16H genetic knockdown via RNAi or pharmacological inhibition by using its inhibitor, curaxin 137 (CBL0137), results in the induction of IFNs and interferon-stimulated genes (ISGs). Through this mechanism, CBL0137 is shown to efficiently inhibit infection of multiple viruses, including Zika, influenza, and SARS-CoV-2. Furthermore, we demonstrated that CBL0137 also causes the remarkable activation of IFN signaling in natural killer (NK) cells, which promotes the NK-mediated killing of virus-infected cells in a co-culture system using human primary NK cells. Overall, our studies unraveled the previously un-appreciated role of FACT complex in regulating IFN signaling in both epithelial and NK cells, and also proposed the novel application of CBL0137 to treat viral infections. Blood glucose level is an important risk factor for the prognosis of infection and critically ill patients. We aimed to investigate the prognostic value of blood glucose level in patients with COVID-19.Research design and methods We collected clinical and survival information of 2041 consecutive hospitalized patients with COVID-19 from two medical centers in Wuhan. Patients without available blood glucose level were excluded. We performed multivariable Cox regression to calculate HRs of blood glucose-associated indexes for the risk of progression to critical cases/mortality among non-critical cases, as well as in-hospital mortality in critical cases. Sensitivity analysis were conducted in patient without diabetes.Results Elevation of admission blood glucose level was an independent risk factor for progression to critical cases/death among non-critical cases (HR=1.30, 95% CI 1.03 to 1.63, p=0.026). Elevation of initial blood glucose level of critical diagnosis was an independent risk factor for in-hospital mortality in critical cases (HR=1.84, 95% CI 1.14 to 2.98, p=0.013). Higher median glucose level during hospital stay or after critical diagnosis (≥6.1 mmol/L) was independently associated with increased risks of progression to critical cases/death among non-critical cases, as well as in-hospital mortality in critical cases. Above results were consistent in the sensitivity analysis in patients without diabetes.Conclusions Elevation of blood glucose level predicted worse outcomes in hospitalized patients with COVID-19. Our findings may provide a simple and practical way to risk stratify COVID-19 inpatients for hierarchical management, particularly where medical resources are in severe shortage during the pandemic. Sendo assim, busca-se avaliar a partir da visão dos sujeitos, a relação das políticas públicas direcionadas ao desenvolvimento da agricultura familiar relacionadas ao paradigma do desenvolvimento rural e à manutenção da atividade no território do Vale do Ribeira – Paraná, mais especificamente nos municípios de Rio Branco do Sul e Itaperuçu, com vistas a verificar a referida contribuição destas políticas públicas no contexto do desenvolvimento rural e cumprimento dos objetivos propostos. Assim, pode-se concluir que a agricultura familiar tem relevância no alcance do desenvolvimento rural e sustentável do meio, mas este êxito é dependente das políticas públicas.
